Abstract

Abstract Background and Objectives: Diarrhea is one of the most common diseases in the world. Campylobacter jejuni is one of the prevalent agents of bacterial diarrhea in most of developing countries. It is usually ignored in routine laboratory test in our country, because it has a difficult investigation method. This article aims to determine the prevalence of Campylobacter jejuni, in diarrhea samples in Gorgan City (East north of Iran) by PCR Method. Material and Methods: This descriptive study was performed on 455 diarrheal samples during one year (2005-06).255 out of them were cultured on Preston media (Himedia co.) on 42°c. DNA Extracted by phenol cholorophorm method was directly carried out on stool samples.16srDNA hipo and asp primers for detection of Campylobacter genus, C.jejuni and C.coli species were used, respectively. In addition, universal primer of 16srDNA was used for control of PCR method. Results: no sample was positive for Campylobacter in culture .only three samples were positive for Campylobacter genus and C.jejuni specific primer but none of them were positive for C.coli .99 samples were positive by universal primer of 16srDNA . Conclusion: The results indicate that C.jejuni isn't a prevalent agent in diarrhea in our region. Key words: Campylobacter jejuni -Gorgan- Diarrhea

Abstract

Abstract Background and objectives: The increase of ESBL producing E.coli can create a tremendous difficult y for the health system. These isolates leads to rapid transmission of causative genes to other clinically important bacteria and synchronously increased resistant to other antibiotics. This study was carried out to determine the prevalence of this isolate and related genes in Gorgan, North of Iran. Material and Methods: This study was conducted on 218 isolated E.coli from urinary tract infection of outpatients referring to six medical laboratories in Gorgan, during 2010-11. The resistance to Cefotaxim (Mast Co.) was assessed by Kirby-Bauer disk diffusion method. The confirmatory test for detection of resistant isolates was carried out by double disk method at the presence of Cefotaxim and clavulanic acid. The presence of β lactamase gene of blatem, blactx and blashv in ESBL was assessed by PCR method. Results: of 218, 70 isolates (32.1%) are resistant to Cefotaxim. Sixty-two (88.6%) of them are confirmed as ESBL producing E.coli. β lactamase genes of blactx, blatem and blashv can be seen in 28(45.2%), 26(41.9%) and 6(9.7%) isolates, respectively. Conclusion: the prevalence of ESBL producing E.coli in Gorgan is in the range of country average and blaCTX-M gene is the most common gene. Key words: E.coli, ESBL, bla gene, UTI

Abstract

Abstract Background and Objective: Given the importance of having knowledge about intestinal parasitic infections in rehabilitation centers, we aimed to investigate the prevalence of intestinal parasitic infections in the disabled. Materials and methods: This cross-sectional study was conducted on 196 stool specimens obtained from four rehabilitation centers in Golestan province, 2009. Triple fecal samples were collected and examined by direct wet Mount, Formalin-ether concentration technique and Trichrome permanent staining (with optical microscope). Results: Of 196 specimens, 24 (12.3 %) had parasitic protozoan infection (5 species) and worm (1 species). The prevalence of intestinal protozoa was 11.8 %, and the highest rate of infection was related to Blastocystishominis (4.2%) and Giardia lamblia (3.1%). The rate of infection was the highest in under 9 and 20-29 years (4.1%) and the lowest in over 40 (0.5%). The prevalence in mentally handicapped was 9.2% and in physically disabled was 2.5%. Conclusion: Due to serious complications of Amoebiasis, Giardiasis and other parasitic infections, it is essential to be careful about personal and social hygiene in these centers. Abstract

Abstract Background and Objective: One of the proteins shared in all strains of Brucella is 31 kDa surface protein (BCPS31) that could be an appropriate target for immunization and serological diagnosis. Material and Methods: In the present study, BCSP31 produced as a recombinant protein in pET28a (+) expression system was utilized, using ELISA, to detect trace specific antibody (IgG) in brucellosis patients' serum that was confirmed by culture. We also evaluated cytokine response of peripheral blood lymphocytes to this protein in the cell culture. Results: The results indicated a significant amount of surface protein antibodies (IgG) in the serum of patients with brucellosis. Evaluation of lymphocyte responses to rBCSP31 also showed a significant IL-12 and IFN-γ production in patients’ lymphocyte cultures. Conclusion: These results suggest that BCSP31 can elicit specific humoral and cellular responses during host infection and it can be used in designing immunization and serologic diagnosis systems. Abstract

Abstract

      Background and Objective: The aim of this study was to determine the chemical composition, antibacterial and antifungal effects of Thymus vulgaris and Cuminum Cyminum essential oils against foodborne pathogens and Candida species in vitro.

      Methods: The essential oils were extracted from the aerial parts of Thymus vulgaris and dried Cuminum Cyminum seeds using a Clevenger apparatus for 3 hours. Analysis of the essential oils’ constituents was performed using gas chromatography-mass spectrophotometry. The antibacterial activity of Cuminum Cyminum essential oil and essential oil of Thymus vulgaris against Bacillus cereus, Listeria monocytogenes, Escherichia coli and Salmonella typhimurium were evaluated in agar culture medium. The minimum inhibitory concentration (MIC) of these essential oils against fungal strains of Candida albicans, C. tropicalis, C. parapsilosis and C. dubliniensis was measured.

      Results: Thymol (64.45%) and cuminaldehyde (29.02%) were the main components of the essential oil of Thymus vulgaris and Cuminum Cyminum, respectively. The largest inhibition zone diameter in the essential oils of Thymus vulgaris and Cuminum Cyminum in the agar disk diffusion method was related to B. cereus with 30 and 21 mm diameter, respectively. The largest growth inhibition zone diameter by the essential oil of Thymus vulgaris in the well diffusion method was 21 mm and against B. cereus. The MIC of essential oil of Thymus vulgaris in the microdilution method was 0.09% against all the four Candida strains. The MIC of Cuminum Cyminum essential oil against strains of C. albicans and C. tropicalis was 0.39%, while it was found as 0.19% against C. parapsilosis and C. dubliniensis.

      Conclusion: In this study, Cuminum Cyminum essential oil and essential oil of Thymus vulgaris show suitable inhibitory effects against the growth of bacteria using well and disk diffusion methods. Regarding the antifungal effects, the MIC of essential oil of Thymus vulgaris is lower than the Cuminum Cyminum essential oil, which indicates the higher antifungal activity of the essential oil of Thymus vulgaris. This study has raised the possibility of using these essential oils as suitable antimicrobial compounds and alternatives for chemical preservatives in the food industry.

Abstract

Background and objectives: Coronary artery disease (CAD) refers to stenosis or obstruction of coronary artery due to atherosclerosis or clotting. The aim of this study was to evaluate possible association of serum miRNA-197 (miR-197) expression as a biomarker for CAD diagnosis.
Methods: In this study, 100 patients with CAD who had angiography and vascular transplantation were selected. Expression of miR-197 was evaluated using real-time RT-PCR technique and the SYBR Green method. The Pearson's correlation coefficient was used to determine relationship of miR-197 expression and severity of coronary artery disease. The t-test was used to determine significance of expression of miR-197 in the study groups. All statistical analyses were carried out in SPSS 16 and at significance of 0.05.
Results: The results showed a direct relationship between miR-197 expression and CAD severity. The relative expression of miR-197 in the CAD patients was significantly higher than that in control subjects (P<0.004).
Conclusion: It seems that miR-197 can be considered as an indicator of coronary endothelial cell function. This microRNA could be used as a biomarker for CAD prognosis and treatment progression.

Abstract

Background and objectives: Bacterial contamination of wounds is a serious problem, particularly in burn patients. Gram-positive bacteria are the predominant cause of infection in newly hospitalized burn cases. This study aimed to survey the prevalence and antibiotic resistance pattern of gram-positive bacterial isolates among burn patients in Rasht, North of Iran.
Methods: This cross-sectional study was conducted on burn patients with a positive culture for gram-positive isolates who were hospitalized in the Velayat Burn Center in Rasht, North of Iran, during 2017-2020. The isolates were identified using standard microbiological methods. Moreover, the antibiotic resistance pattern was determined by the disk diffusion method.
Results: During the study period, 671 bacterial cultures were obtained, of which a total of 16 gram-positive isolates were taken from the patients. The frequency of coagulase-negative staphylococci (CoNS), Staphylococcus aureus, and Enterococcus spp. was 68.7%, 18.8%, and 12.5%, respectively. In addition, the highest rate of resistance in CoNS isolates was against trimethoprim/sulfamethoxazole. The highest rate of resistant among S. aureus isolates was recorded against penicillin. Moreover, Enterococcus faecalis isolates showed a high level of resistance to ampicillin, erythromycin, tetracycline, gentamicin, and ciprofloxacin. All isolates were susceptible to teicoplanin. Moreover, the frequency of methicillin-resistant S. aureus isolates was 66.7%.
Conclusion: Given the increasing prevalence of drug-resistant strains, especially in susceptible burn patients, it is imperative to analyze the bacterial etiology of nosocomial infections periodically and epidemiologically.
