Abstract

Abstract Background and Objective: The high occurrence of early childhood caries (ECC) is one of the most common problems in children dentistry. Streptococcus mutans and lactobacilli.spp are closely associated with the development of early childhood caries (ECC). Therefore, the aim of this study was to investigate the frequency of streptococcus mutans and lactobacilli.spp in 3-5 –year- old children with and without dental caries. Material and Methods: This case-control study was conducted on 60 children aged 3 -5-years, without any history of systemic disease, who had not received any antibiotic therapy and fluoride usage during the last month. The cases were divided into three groups of early childhood caries, usual caries and caries-free. The infected dentin samples were collected from cervical and proximal in ECC and usual caries group, respectively. Also in all the three groups, the samples of dental plaque from buccal surfaces were collected and immediately immersed into Brain Heart Infusion (BHI) broth medium. After that, the diluted sample was plated onto MitisSalivarius agar (Difco) for detecting streptococcus mutans and Rogosa agar (Difco) for detecting lactobacilli.spp. Data were analyzed by Chi- Square and ANOVA. Results: of the samples taken from dental plaque, S. mutans is observed in 90% of ECC, 80% of proxymolcarries and in 25% of caries-free individuals. Based on the results, the presence of S.mutans in the group of caries is significantly higher than that of without caries, but there is no significant difference between the two groups of caries. In addition, lactobacill.spp is isolated from 60% ofthe samples, but there is no significant difference between two groups of caries and caries-free. The average of decayed, missing and filled teeth (DMFT) index in the group with early childhood caries (6.95±1.572) and usual caries (5.80±1.105) is different significantly. Pearson correlation test shows that there is a significant relationship between caries experience and the amount of S.mutans, but there isn’t this relationship withlactobacill.spp. Conclusion: The findings show that the children with early decay have the higher level of S.mutans in their dental plaque. The most common strain isolated from dentin, without consideration of their location, is S.mutans. Key words: Early dental care, Dental caries, DMFT index, Streptococcus mutans, Lactobacilli.spp.

Abstract

Abstract Background and Objective: Allergic rhinitis can be stimulated by several allergens. Molds are among these allergens and it is important to assess their frequency in different geographic area. Hence, we aimed at determining the frequency of mold allergens in allergic rhinitis patients referred to specialized clinics of Tabriz Imam Reza hospital, 2011. Material and Methods: This cross-sectional study was conducted on the serums of 90 rhinitis patients diagnosed by specialized physician. Using Immunoblotting method, the level of specific IgE against four molds including Penicillium, Aspergillus, Alternaria and Cladosporium were investigated. Results: Of 90 Patients, 40 were men (44.4%) and 50 were women (55.6%). The participants were between 6 to 53 years and the most were 28-31years. The allergy was related to Penicillium (3.3%), Aspergillus (5.6%), Alternaria (13.3%) and Cladosporium (4.4%). There was a significant statistical relation between age and allergic rhinitis to Alternaria (P=0.011). Conclusion: Molds can grow and proliferate in very humid environments. Because of low humidity climate in Tabriz (in the northwest of Iran), allergy to molds is relatively low in this region. Key words: Rhinitis Allergic Mold Allergy

Abstract

Abstract Background and Objective: Burkholderia cepacia complex (BCC) is a plant pathogen that is an important mortality factor in immune-compromised and hospitalized patients. We aimed to Isolate and Characterize the Burkholderia Cepacia Strains from Hospitalized Patients in the Hospitals of West Guilan Province. Material and Methods: This study was conducted on 90 saliva and blood samples obtained from patients with blood infection, pneumonia, asthma, patients connected to the monitoring and ventilator systems, and immune-compromised patients in different sections of hospitals such as the pediatrics, internal section, ICU and CCU. Primary screening was performed by cultivating the samples on Burkholderia cepacia selective agar (BCSA) Sensitivity to antibiotics was tested by Kirby-Bauer and Muller-Hinton Agar (MHA) and the separated isolations were recognized by strengthening the gene rec A. Results: Of 90 isolated samples, only one strain suspected B. cepacia was isolated from 24-year old women with asthma. This strain was resistant to bacitracin, pipracillin and ciprofloxacin antibiotics. Conclusion: The incidence of B. cepacia is rare in western part of Guilan province, which is congruent with the results of overall incidence in Iran.

Abstract

ABSTRACT
            Background and Objectives: Formetanate hydrochloride is an effective insecticide and acaricide, which is frequently used in citrus gardens. Given its high toxicity, it is necessary to evaluate effects of this insecticide on living factors. In this study, we investigate effects of formetanate hydrochloride on human hemoglobin using spectroscopic analysis and molecular docking modeling.
            Methods: Effects of formetanate hydrochloride on human hemoglobin were investigated using molecular docking technique and thermodynamic and spectrophotometric methods such as normal and synchronous fluorescence spectroscopy, UV-Vis absorption spectroscopy and circular dichroism.
            Results: Decrement of the soret band without any significant shift proved that the insecticide could penetrate into the heme pocket and directly interact with the porphyrin ring. Results obtained from the intrinsic fluorescence spectroscopy and heme degradation study at 321 and 460 nm indicated that formetanate hydrochloride could potentially destroy the heme prosthetic group and eliminate its filtering effect, which results in fluorescence enhancement. The fluorescence study demonstrated a competitive behavior between hydrophobic probe ANS and formetanate hydrochloride for binding to the hemoglobin hydrophobic patches. Results of circular dichroism and synchronous fluorescence spectroscopy confirmed the structural change and polarity alteration around aromatic fluorophores.
            Conclusion: Our findings elucidate that the binding of formetanate hydrochloride to hemoglobin through the heme pocket is energetically much more favorable compared to binding to the hydrophobic central cavity. In addition, formetanate hydrochloride can cause conformational changes in the human hemoglobin and the competitive behavior of this insecticide for the oxygen site can subsequently reduce the oxy form of hemoglobin.
