Abstract

Abstract Background and objectives: Germ tube-producing ability of C.albicans in human serum is one of the most important virulent factors inducing transformation yeast to mycelia form. This phenomenon can be altered by some environmental and nutritional factors. The purpose of this study was to investigate the effect of temperature, pH and glucose concentrations in germ tube formation of C. albicans in an in vitro system. Material and Methods: The germ tube production test in human serum (with normal glucose titer) in different temperature, pH, and glucose concentrations was conducted invitro using standard strain of C. albicans (ATCC 10231). The average number of cells with germ tube after 2 hours and the rate of yeastmycelial transition were analyzed using one-way ANOVA test. Results: Maximum germ tube production rate is seen in temperature of 37°C, pH of 6.5 and glucose concentration of 30 mg/ml (P= 0.0001) and also germ tube is seen in earliest time in these conditions in invitro. Conclusion: It seems that these environmental and nutritional factors in human body particularly in diabetics can make this fungus to produce germ tube and invade to the different tissues. Abstract

Abstract Background and objectives: Air ionization (AI) may reduce the microbial content of the air 'and has bactericidal effects on some bacterial Strains, which is controversial. We conducted this study to evaluate the efficacy of Air ionizer in reducing the microbial content of air. Material and Methods: This experimental Study was carried out in Sadi Hospital of Isfahan. Two air ionizer were placed in two boxes. and the third box (Control) was empty. At the beginning of experiment and every 30 minutes up to three hours, we did air sampling in all groups. After taking the samples of every box, we incubated the samples and then performed colony counts and bacteriologic studies. Results: In all thirty-minute intervals, Colony Counts in all groups were lower than control group (P<0.05). 'Coagulase negative staphylococcus (CNS) was the most common bacteria isolated followed by Bacillus spp, Acinetobacter and Escherichia Coli, in control group, no colonization of Acinetobacter and E.coli is found in Air ionizer groups. Conclusion:Our findings qualitatively indicate that air ionization can reduce the microbial content of the air. Regarding the type of microbial air pollution and the amount of air cleaning needed, this method can be used solely or in combination with other air cleaning methods. Abstract

Abstract Background and objectives: Pseudomonas aeruginosa as an opportunistic pathogen can establish lethal infections in immunocompromised patients or those exposed to predisposing factors. This bacterium contains a single polar flagellum causing motility, chemotaxis and colonization in acute phase of infection. The flagella filament is made up of a structural protein called flagellin. This study was aimed at determining The frequency of fliC gene in Clinical Samples. Material and Methods: In this study, a pair of specific primer for types of flagellin (a, b type) was designed and by using PCR method its structural gene (fliC) was recognized and amplified in clinical strains. Results: This original primer has appropriate efficiency in diagnostic of pseudomonas aeroginosa flagellum. Our study shows that 85% of the Clinical Samples have a fliC gene. Conclusion: This method can be applied to recognizing of the motile strains, and their antigenic typing, and complete amplification of fliC sequence in order to cloning and expression of recombinant flagellin. Abstract

Abstract Background and objectives: cardiac surgery is often associated with acute kidney injury (AKI). Nowadays, AKI is typically diagnosed by an increase in serum creatinine, which is a delayed and unreliable biomarker. Recent studies recommended using the liver type fatty acid binding protein (L-FABP) as an early biomarker. Material and Methods: The urine samples of 18 adult patients undergoing cardiac surgery were collected in different times before (2, 4,8,24 hour) and after cardiac surgery for detection of L-FABP by Elisa. Results: The results from ELISA test show that the increasing amount of L-FABP in urine samples of 4 patients is a diagnostic indicator for AKI. The mean concentration of L-FABP has increased up to 17 times at 8 hours after cardiac surgery compared to before surgery. Conclusion: according to our findings, we speculated that the urinary L-FABP can be a reliable and rapid biomarker for diagnosis of acute kidney injury. Abstract

Abstract Background and objectives: In this research, the formation of chitosan-TiO2 nanocomposite and its antibacterial effect on Escherichia coli and staphylococcus aureus was investigated Material and Methods: to study the results, we used Scanning electron microscopy (SEM) and transition electron microscopy (TEM) images, infrared (IR) spectroscopy and ultraviolet-visible. Optical Density (OD) was also measured by spectrophotometer then the effect of this nano composite, in the vicinity of aforementioned bacteria, on the sterilized gauze in solid Muller Hinton Agar and TSB liquid mediums was assessed Results: The mentioned nanocomposite was formed with the composition of 4mg/ml Chitosan concentration and 2% titanium dioxide concentration. Finally, we observed that this nanocomposite near 100% could prevent bacterial growth and in the presence of this material did not grow any bacteria. Conclusion: chitosan-Tio2 Nanocomposite can be useful on culture medium and sterilized gauze to control pathologic bacteria. Abstract

Abstract Background and Objective: Pre-eclampsia is the most common cause of maternal and neonatal mortality. Recent studies show that there may be a relationship between chorionic gonadotropine (HCG) and pre-eclampsia. Thus we aimed at assessing the serum level of βhCG after 28th week of pregnancy in mild pre-eclamptic and healthy pregnant women. Material and Methods: In this case-control study, we measured the level of βhCG in 26 pre-eclamptic and 26 normal pregnant women. The difference between groups was analyzed by Mann-Whitney test (P≤ 0.05). Results: The mean of βhCG in the case (73192 ± 42956 IU/L) and control group (34038 ± 21272 IU/L) is different significantly (P= 0.001). Considering mother’s age, gestational age and parity, there is a significant difference between the two groups (P=0.001). Conclusion: The results of our study show that the higher level of βhCG, the greater the risk of pre-eclampsia. Key words: Pre eclampsia, βhCG, Parity

Abstract

Abstract Background and Objective: Helicobacterpylorus is the major cause ofinflammation andulcer instomach, and immunoglobulin IgG is one of the antibodies produced against it, which is important in the course and diagnosis of the previoussufferers. The awareness of the prevalence of this disease can be helpful for the physicians to choose the way of treatment. Material and Methods: In these cross-sectional study, 516patients referred to laboratory was studied. After separating the serum, Antibody Helicobacter pylori IgGtest wasdone by ELISA method. Results: of 516, 156 (30.2 %) of the patients have a positive result, 51(32.7%) are males and 105 (67.3 %) are females. Positive percent of males (43.5%) is greater than females (26.5%).Over-45-year-old women (9.8 %) have the highest percentage of disease titers. Conclusion: The percent of positive cases in men is more than that the women. Over-45-year-old women (9.8 %) have the highest percentage of positive case. Abstract

Abstract Background and Objective: Nowadays, nosocomial infection is one of the greatest problems in hospitals. Normal flora of staff’s hands and the bacterial agents on the surface of medical equipment can become progressively colonized with potential pathogens during patient care. This study was carried out to determine the bacterial agents existed on staff’s hands and in the wards of hospital to step in to control nosocomial infection. Material and Methods: In this descriptive study, during 17 months (22.mar.2010- 30.aug.2011), 403 samples, using sterile swab , were randomly obtained from the staff’s hands and medical equipment of emergency departments , ICU, male operation room and female surgical unit . The samples were cultured on Blood agar (BA) and Eosin methylene blue (EMB). Then, identification of isolated bacteria was done with diagnostic tests. Results: Of 430 samples, 530 bacteria were isolated from staff’s hands (N= 291) and medical equipment (N= 234). The most common bacterium from personnel’s hands (144 49.5%) and medical equipment (24 10%) is Staphylococcus aureus. Also, three isolates of pseudomonas aeruginosa from staff’s hands of male surgical ward and medical equipment of ICU, and two isolates of Acinetobacter.spp from ICU’s medical equipment were identified. Conclusion: With regard to the findings, it seems that applying the appropriate disinfectant agents by using standard procedures is necessary. Abstract

Abstract Background and Objective: Of the most common hypochromic microcytic anemia are iron deficiency anemia and minor thalassemia, which are common in Iran and their differential diagnosis is extremely important. The level of 25-hydroxy vitamin D is the indication of vitamin D blood status. The aim of this study was to compare serum levels of vitamin D in people with minor thalassemia and iron deficiency anemia with healthy subjects in order to investigate the relationship between vitamin D deficiency and iron absorption. Material and Methods: In this case-control study, 24 patients with minor thalassemia, 20 patients with iron deficiency anemia and 24 healthy individuals participated. Groups were matched for age and sex. Testing of Vitamin D level by ELISA, ferritin by quantitative luminescence method and HbA2 by column chromatography was carried out. Results: The number of individuals with low level of vitamin D in iron deficiency group is 15 (75%), in minor thalassemia group is 8 (33/3%) and in the control group 11 (45.8%). Conclusion: In this study, the highest percentage of vitamin D deficiency is observed in cases with iron deficiency anemia. Because of association between vitamin D and anemia, iron and vitamin D supplementation is recommended to enrich the diet. Keywords: Anemia Minor Thalassemia Iron Deficiency Anemia Vitamin D

Abstract

Abstract Background and Objective: In many countries, especially in developing countries, diarrheal diseases is one of the most important causes of mortality. In these countries, due to availability transfer conditions of parasitic infections, (pathogenic and non-pathogenic parasite), have a high prevalence. This study aimed was determining the prevalence of Parasites causing diarrhea in Gorgan . Material and Methods: This cross-sectional study was performed on 1086 Patients with acute diarrhea. The test was performing by the wetmount and formalin-ether concentration method according to WHO protocol. Results: From 1086 patients in study, 197 (18.1%) were infected by intestinal parasites, that 141 (13%) were infected with Protozoan and 79 (7.27%) were infected with intestinal worms. The prevalence of wet mount method was, Entamoebahistolytica in 69 (6.4%), Giardia lamblia in 27 (2.5%), Entamoeba coli in 15 (1.4%), Chilomastixmesnili in 8 (0.7%), and with formalin-ether method was Hymenolepis nana in 26 (2.4%), Trichostrongylus in 16 (1.5%), Enterobiusvermicularis in 10 (0.9%). Abstract

Abstract Background and Objective: Microsporidia is an obligatory intracellular parasite known as an opportunistic infection in immunocompromised patients. Its laboratory diagnosis is relatively difficult and modified trichrome staining is a standard diagnostic method for detection of Microsporidia. The aim of present study was to identify intestinal Microsporidia in kidney- transplanted patients, using modified Trichrome Staining and Calcofluor White Methods. Material and Methods: In 2012, 180 stool specimens were taken from kidney transplanted- patients given immunosuppressive drugs in Hamadan, Iran. To identify Microsporidia we use modified Trichrome Staining and Calcofluor White Methods. Results: The mean duration of kidney transplant and immunosuppressive drug using was 5.5 Years. Only one female patient was positive for Microsporidia. Conclusions: Owing to low frequency of this opportunistic infection among kidney transplanted patients, we can conclude that their hygienic conditions are good enough and they are not exposed with the parasites. Abstract

Abstract Background and Objective: Laboratory personnel have always accidental exposure to clinical samples, which can cause the transmission of infection. This threat can be prevented and controlled by education for the use of safety instruments. The purpose was to determine the frequency of accidental exposure to laboratory samples among Yazd laboratory personnel in 2011. Material and Methods: This descriptive cross-sectional study was conducted on 100 of Yazd clinical laboratory personnel. The data was collected, using a valid and reliable questioner, via interview and analyzed by means of SPSS software. Results: Eighty-six percent of the subjects reported an experience of accidental exposure to clinical samples, such as blood, serum and urine. The causes were carelessness (41%) and work overload (29%). Needle- stick was the most prevalent injury (52%) particularly in sampler workers (51%) and in their hands (69%). There wasn’t significant relationship between accidental exposure to laboratory samples and the variables such as private and governmental laboratories (p=0.517), kind of employment (p=0.411), record of services (p=0.439) and academic degree (p=0.454). The subjects aged 20-29 (p=0.034) and worked in sampling unit had the highest accidental exposure. Conclusion: Based on the results, inexperience of the personnel especially in sampling room, overload at work and ignorance of applying safety instruments are known as the most important reasons for accidental exposure to clinical samples. Abstract

Abstract Background and Objective: Chitin, which is a linear polymer of N-acetyl glucosamine residues, has been the most abundant polymer in nature after cellulose. In recent decades, Chitinases have received increased attention because of their wide range of applications, especially in biological control against fungi. Material and Methods: the isolation of bacilli producing chitinolytic enzymes was performed by collecting 40 soil samples from various regions of Gorgan, northern of Iran. The chitinolytic potential of the isolates was indicated by observation of clear zone in colloidal chitin agar medium. Identification of selected strains was performed by polyphasic taxonomy, and subtler identification and sequensing were carried out by extraction DNA. Antifungal effect was evaluated by well method against Candida albicans (ATCC 10231) Aspergillusniger (ATCC 2029)،Aspergillusflavus (IR6) Fusariumoxyporum (PTCC 5115) and Alternariaalternata (PTCC 5224). Results: Nine colonies of chitinase positive bacillus were isolated on choloidal Chitin Agar (CCA) and five of them had antifungal effect. R6 strain had the highest, and R2 and R3 had the lowest effect on fungi. The 16S rRNA sequence of these isolations in comparison with the known bacteria has 95-97% similarity. Conclusion: Some of the soil bacteria can have antagonestic effects on human and phytopathogenic agents existed in soil. Keywords: Bacillus Chitinase Soil Antifungal

Abstract

Abstract Background and Objective: Stenotrphomonas maltophilia is an opportunistic nosocomial pathogen with high mortality in immunocompromised cases. The aim of this study was to isolate and identify Stenotrphomonas maltophilia in the hospitals’ environment and wards. Material and Methods: In this cross-sectional study, a total of 1108 samples were collected from environment of two hospitals during 12 months. Identification of isolates was performed using biochemical, phenotypic (intrinsic resistance to carbapenems) and molecular methods (amplification of 23S rRNA gene). Results: Of the studied samples, 186 (16.78%) nonfermentative gram negative bacilli (NFGNB) were identified. Amongst NFGNB, 18 (1.62%) isolates were identified as S. maltophiliaby using biochemical tests. Of 18 biochemically identified isolates, 15 (83.3%) were confirmed via PCR. Sinks (40%) and men surgery ward ( 33.3 %( were the most contaminated sites and wards of hospitals, respectively. Conclusion: S. maltophilia is repeatedly isolated from sink which shows that the moist hospital environments need to be considered as a source for dissemination of bacteria. Abstract

Abstract Background and Objective: Honey is a healthy and nutritious food that has been used for a long time as a treatment for different diseases. One of the applied properties of honey is its antimicrobial effect, which differs between different types of honey due to variation of phenolic and antioxidant compositions. This study aimed to assess antimicrobial effect of honey on Bacillus cereus, considering its chemical properties. Material and Methods: Three samples of honey (A1 and A2 of Khorasan Razavi Province and A3 of South Khorasan province (were prepared and studied in terms of chemical parameters .The antibacterial effect of honey was surveyed throughTurbidimeter using spectrometer with incubator time of 2, 4, 6, and 8hrs. the level of turbidity caused by bacterium growth was measured at different times with a wavelength of 600nm. Results: According to the study, the samples containing higher concentration of polyphenol has more antimicrobial activity. The samples of A2, A3, and A1 had the highest concentration of polyphenol, respectively. Conclusion: The results indicate the prebiotic effect of honey that can be justified by the presence of fructo-oligosacharids and vitamin B. Abstract

Abstract Background and Objective: The hormonal responses are different, according to type, intensity and the duration of training. We aimed to compare the effect of endurance and resistance training in untrained men on the level sexual hormone including testosterone, estradiol, and on sex hormone binding globulin (SHBG). Material and Methods: this study was conducted on male Student Volunteers divided randomly into three groups of eight members, including endurance, resistance and control. To measure the variables, we performed sampling before and after the training program. Results: the levels of testosterone and estradiol, in both groups have a significant (p<0.05) increase and decrease respectively. SHBG level was significantly increased only in endurance group (p<0.05), But LH level was meaningfully increased only in the resistance group (p<0.05). Conclusion: The endurance and resistance training can increase the production of testosterone .Increased testosterone and decreased estradiol in men lead to sperm production and improve fertility. Abstract

Abstract Background and Objective: Vancomycin is used for treatment of methicillin-resistant S. Aureus (MRSA) infections therefore, resistance to this antibiotic is increasing. We aimed to determine the antibiotic resistance pattern and frequency of vancomycin resistant S. Areas (VRSA) strains isolated from clinical samples. Material and Methods: In this cross-sectional study, 100 S. Aureus isolates collected from hospitals in Shiraz during six months, 2012, were identified by biochemical, microbiological and molecular methods. After determination of antibiotic susceptibility pattern by disc diffusion method and vancomycin agar screening test, Minimum Inhibitory Concentration (MIC) was determined by E-test for vancomycin, thicoplanin, linezolid and quinupristin-dalfopristin. Results: The most resistant and the most sensitive antibiotic were ampicillin (%95) and quinupristin-dalfopristin (99%), respectively, and 44% of isolates were resistant to methicillin. In agar screening test, 48% of strains had reduced sensitivity and in disc diffusion 3% strains were resistant to vancomycin. In E-test method, only one isolate was resistant to vancomycin. Conclusion: given the presence of VRSA and new antibiotic resistant strains, we recommend doing some intervention to prevent from spreading these strains in hospitals. . Abstract

Abstract Background and Objective: Diarrheal diseases may occur in all age groups, worldwide. Escherichia coli (E. coli O157: H7) is one of the most important bacterial agents causing this disease. The purpose of this study was to assess the prevalence of diarrhea in patients with acute diarrhea caused by this bacterium. Material and Methods: In this study, 214 stool samples were collected from acute diarrheic patients in hospitals and clinical laboratories of Malayer city, Iran. The samples were cultured in rich, differentiation, specific and selective medium. To detect E. coli O157: H7, rapid detection method and antisera were used . Results: In 12.15% of the samples, lactose fermentation was observed and E. coli was afirmed. The results of lactose positive samples showed that 6.54% were negative-β- Glucuronidases and were confirmed as E. coli O157. Finally, by using O157: H7 antisera, 1.87% of 14 samples that were negative- β-Glucuronidases were asserted as E. coli O157. Conclusion: based on the results, the prevalence of E. coli O157: H7 is higher in children and elders than the others. Food and water play a significant role in transferring E. coli. Abstract

Abstract Background and Objective: Efficient human resource is the major advantage of an organization over others. Commitment to organization not only may reduce sick leave, delays and displacements but also give rise to a significant increase in productivity, more vitality among staff, better attainment of the organizations' goals and personal aims. This study aimed to evaluate the relationship between personal characteristics and organizational commitment among medical laboratory staff. Material and Methods: This correlational study was conducted on 105 medical laboratory staff selected randomly in Gorgan, 2010-2011. The instruments for data collection were NEO personality inventory and Meyer and Allen's model of organizational commitment. Results: The relationship between organizational commitment and the variables such as neuroticism, openness, agreeableness and Conscientiousness was positively significant but that was not the case for extroversion characteristic. Conclusion: Given the positive relationship between personality characteristics and organizational commitment, we recommend improving these characteristics to enhance staff commitment to organization.
