Abstract

Background & objective:

One of the common purposes of medical research is Determination of effective factors on the occurrence of event. Due to the interaction of risk factors regression models, discriminant analysis and classification procedures used. Uses of these models require making the assumption which in the medical data isn’t usually established. Therefore, alternative methods must be used. According to diversification of risk factors for of esophageal cancer, the purpose of this article is the Introduction and application of classification and regression tree for determination of risk factor for esophageal cancer in Golestan province.

Methods:

Data of this article gathered from case-control study. Case group contain all confirmed cases of esophageal cancer that consist of 90 male and 60 female subjects in Golestan province during one year. Two control groups were considered for each case. Control groups were selected from family of patients and neighbors and matched for age, sex, ethnic and place of residence. Data was analyzed with classification and regression tree model and by using of R software. Gini criterion was used for selection of best splitting in each node and ROC surveyed accuracy of CRT model.

Results:

(ethnic factors) can be effective in esophageal cancer occurrences.

Results of Classification tree model showed that exposure to CT and X-ray dye (socio-environmental factors), unwashed hands after defecation, history of smoking (lifestyle factors) and family history of cancer

Conclusion:

models results` interpretation are two essential beneficiary of these models which can use in medical sciences.

Abstract

Background and objectives:

Fatigue experience is a psychological experience therefore, the relevant caregivers’ acquaintance with this issue

is the most important step in offering optimal care for the patients. Accordingly, this study was conducted

to determine Cancer-related Fatigue and its Relationship with Demographic and Clinical Characteristics in

Cancer Patients.

A cancer diagnosis is a highly undesirable event for anyone.

Methods:

(P=0.018), the treatment regimen type (P<0.001), and the family’s economic status (P<0.000) were found to

The patients’ mean age was 48.39±1.5 Of the patients, 55.3% had undergone less than 12 months

have a significant relationship with fatigue.

Conclusion:

Psychological interventions, besides physical caring and nursing interventions play a significant

role in the all-inclusive management of cancer patients’ problems, in particular, their fatigue.

Abstract

Background & Objectives: Breast cancer is the most common cancer among women and the second most common cause of cancer death. Genetic factors play an important role in the development of breast cancer. Among these genetic factors, CHEk2 (checkpoint kinase 2) gene, as a tumor suppressor gene, plays a critical role in DNA repair. Germline mutations in CEHK2 result in the loss of this feature. One of the mutations in CHEK2 gene is a 5395 bp deletion mutation which has been associated with the increasing risk of Breast Cancer in some populations in the world.  In the present study, we investigated the association between a 5395 bp deletion mutation in CHEK2 gene and the risk of Breast Cancer in the women of an Iranian population.

Methods: Pathologic information of 38 cases under the age of 45 and 62 cases over the age of 45 referring to surgery ward of Milad Hospital in Tehran were extracted. 100 healthy controls were included in the study as well. After obtaining informed consent, 5 mL whole blood was taken DNA was successfully isolated. Multiplex PCR was used to investigate the association between a 5395bp deletion mutation in CHEK2 gene and increasing risk of Breast Cancer among patients.

Results: The 5395bp deletion mutation in CHEK2 gene was not found in any of the participating groups of patients or heathy controls.

Conclusion: The present study revealed that there is no significant relation between increasing the risk of Breast Cancer and bearing large deletion mutation in exon 9 and exon 10 of CHECK2 gene.

Abstract

Background & Objective: Inability to measure exact exposure in epidemiological studies is a common problem in many studies, especially cross-sectional studies. Depending on the extent of misclassification, results may be affected. Existing methods for solving this problem require a lot of time and money and it is not practical for some of the exposures. Recently, new methods have been proposed in 1:1 matched case–control studies that have solved these problems to some extent. In the present study we have aimed to extend the existing Bayesian method to adjust for misclassification in matched case–control Studies with 1:2 matching.

Methods: Here, the standard Dirichlet prior distribution for a multinomial model was extended to allow the data of exposure–disease (OR) parameter to be imported into the model excluding other parameters. Information that exist in literature about association between exposure and disease were used as prior information about OR. In order to correct the misclassification Sensitivity Analysis was accomplished and the results were obtained under three Bayesian Methods.

Results: The results of naïve Bayesian model were similar to the classic model. The second Bayesian model by employing prior information about the OR, was heavily affected by these information.

The third proposed model provides maximum bias adjustment for the risk of heavy metals, smoking and drug abuse. This model showed that heavy metals are not an important risk factor although raw model (logistic regression Classic) detected this exposure as an influencing factor on the incidence of lung cancer. Sensitivity analysis showed that third model is robust regarding to different levels of Sensitivity and Specificity.

Conclusion: The present study showed that although in most of exposures the results of the second and third model were similar but the proposed model would be able to correct the misclassification to some extent.

Abstract

Background and objectives: Deregulation in the expression of microRNAs is involved in the pathogenesis of various malignancies. Impaired microRNAs processing pathway is one possible mechanism for global deregulation of the miRNAs. Exportin 5 (XPO5) is a key member of this pathway that links nuclear and cytoplasmic steps of miRNAs biogenesis together. XPO5 deregulation has been reported in some cancers but very little is known about its role in breast cancer. Therefore, this study aimed to evaluate the mRNA expression of XPO5 in breast cancer in an Iranian population.

Methods: In this case-control study, 30 tumoral tissues and 30 tumor-free margins were collected from breast cancer patients. After RNA extraction and cDNA synthesis, XPO5 mRNA expression level was assessed using quantitative Real-Time PCR.

Results: Our results showed that XPO5 was overexpressed in 53.3% of tumoral tissues but the difference in the gene expression level between tumoral tissues and tumor-free margins was not statistically significant (P-value=0.834). XPO5 expression level showed no statistically significant correlation and association with clinical and pathological parameters.

Conclusion: Overexpression of XPO5 in large percent of patients indicates that high level of XPO5 expression may be a tumorigenic factor for breast cancer which needs to be investigated more deeply.

Abstract

Background and Objectives: Human papilloma virus (HPV) is known as the etiologic agent of cervical cancer and second common cancer among women. HPV viruses with the elevated risk of infection have more potentiality to cause cancer. The carcinogenesis in these viruses is accomplished by oncoproteins such as E7. Employing DNA vaccines which code specific antigens such as E7 is a novel therapeutic approach against such cancers.

Methods: In the present study, plasmid coding HPV16 E7 was administered intracutaneously to C57BL/6 tumoric mice models for investigation of its immunostimulating potential. PcDNA3.1+ vector was used as control vector. After immunization, spleen of animals were removed. Then, release of lactate dehydrogenase (LDH) was evaluated to address the cytotoxic activity (CTL) induced by cellular immunity in spleenocytes. Interferon-γ (IFN-γ) and interleukin-4 (IL-4) cytokines were also analyzed as profiles of Th1 and Th2, respectively. Anti-inflammatory cytokine interleukin-10 (IL-10) levels were also investigated in tumor microenvironments.

Results: Our results showed that CTL activity was higher among samples receiving HPV16 E7 coding vector in comparison to the group receiving pcDNA3.1+ control vector (P < 0.05). Levels of IFN-γ and IL-4 were also higher in the group receiving HPV16 E7 plasmid in comparison to the control group (P < 0.05). Similarly, IL-10 levels were significantly lower in tumor carrying mice groups receiving HPV16 DNA vaccine compare to PBS and pcDNA3.1 receiving control groups.

Conclusion: HPV16 E7 expressing DNA vaccine could increase the release of LDH due to immune system CTL activity. Elevation in IFN-γ and IL-4 levels as well as IL-10 reduction indicates an increase in both Th1 and Th2 profiles resulted by using potent DNA vaccine coding HPV16 E7 in tumor animal model.

Abstract

Background and Objectives:  Cervical cancer is one of the most common types of genital cancer among women in developing countries. Addicted women are among the high-risk group of patients who suffer from cervical cancer. Health Protection Motivation Theory (PMT) is one of the theories that emphasizes on fear effects of health risks (such as diseases) on attitudes and health behaviors. Thus, this study was performed to determine the factors associated with cervical cancer screening behavior using PMT on addicted women referring to addiction treatment center of Hamedan in 2015.

Methods: The current study was conducted on 56 addicted women referring to Hannah's Methadone Therapy Counseling Center in June 2015. A researcher-made questionnaire which comprised of three sections (demographics, knowledge and assessment of the PMT constructs) was used to collect data.

The validity of questionnaire was evaluated using content validity method and test-retest test was used to measure reliability. Data were analyzed statistically by SPSS version 16 and using T-test, Chi-Square and ANOVA tests.

Results: The mean age of all participants was 46.48±10.82 years. The average duration of addiction was 8.11±7.22 years. Of all samples, 53.6% had never done any Pap smear test. There was not a  significant relevance between test performance (screening method) and the marital status, income or the last pregnancy method. However, there was a significant relationship between test performance and education (P=0.041), awareness level (P<0.001), structures of motivation (P<0.001), perceived costs (P=0.031), perceived rewards (P=0.001), self-efficacy (P<0.001) and efficient response (P<0.001).

Conclusion: We showed that there was a significant relevance between test performance and the level of awareness and various domains of PMT. Thus, it is recommended that PMT could be used during educational interventions and counseling procedures.

Abstract

Background and objectives: Prostate cancer is the second leading cause of mortality among men worldwide, and the eighth cancer-related cause of death in Iran. Male sex hormones (androgens) are the main cause of prostate cancer cells. Triptorelin is considered a synthetic decapeptide analogue of gonadotropin-releasing hormone (GnRH) in the treatment of prostate cancer. This study aimed to evaluate the methods to deal with the constraints in use of lipid nanoarchaeosomes. Due to their adjuvant feature and presence of ether links, archaeosomes are more stable in blood, compared to other lipid-based formulations. In the present study, the effects of nanoarchaeosomes containing triptorelin acetate on the cellular toxicity of the PC3 prostate cancer cell line were evaluated.
Methods: Halobacterium salinarum bacteria were cultured in HS medium, and their polar lipids of the membrane were removed applying the Blight&Dye technique in order to obtain nanoarchaeosome containing membrane polar lipids. In the next stage, nanoarchaeosomes were prepared by the hydration of polar lipids. In addition, cell viability was evaluated using the MTT assay on the PC3 cell line.
Results: In this research, mean diameter of the drug-containing nanoarchaeosomes was estimated at 263 nm using the ZetaSizer device. Moreover, drug loading efficiency of nanoarchaeosomes was estimated at 98%. Furthermore, the toxicity of the free and nanoarchaeosome drugs was assessed via the MTT assay, results of which were indicative of a mean of IC 50 0.22 µg/ml in this regard.
Conclusion: According to the results of the study, archaeosomes extracted from polar lipids were produced with high concentration and purity on the nanoscale. Considering the high stability, ease of production, level of drug loading, and toxic effect on cancer cells, nanoarchaeosomes can be used as a suitable technique for targeted drug delivery in future studies.

Abstract

Background and objectives: Breast carcinoma is one of the most common malignant diseases among women worldwide. In Iraq there are noticeable elevation in incidence rates and prevalence of advanced stages of breast cancer. Cobalamin (vitamin B12) is essential micronutrient involved in one carbon metabolism and DNA methylation, which affects cancer. All of these may be change in breast cancer. The present work was designed to estimate and compare serum vitamin B12 among female breast cancer patients (60 ones) and healthy control subjects (60 ones) in Sulaimania city.
Methods: This is a case-control study conducted on sixty cases of newly diagnosed women with breast cancer, the control group include sixty healthy women. Serum vitamin B12 levels were estimated by electrochemiluminescence immunoassay (Elecsys) method. Data was analyzed using the software SPSS (Ver. 22) including frequency and percentage for categorical variables. Pearson chi-square test was used for analysis of all categorical variables.
Results: In this study we found that serum vitamin B12 levels were significantly (p=0.01) lower in breast cancer patients as compared to healthy control subject. There was no association between serum vitamin B12 levels with estrogen, progesterone, and HER2 receptor.
Conclusion: Given the results, it can be concluded that serum vitamin B12 is consistently lower among breast cancer patients. There was no association between serum vitamin B12 levels and hormones receptors status, indicating clinical implications for the interpretation of serum vitamin B12 levels. Therefore, it should be taken into consideration by physicians and cancer specialists.

Abstract

Background and objectives: patients undergoing Chemotherapy are severely susceptible to infections due to a compromised immune system and also their oral cavity is a great place for microorganisms and fungi to grow. The aim of this study was to determine the distribution of different strains of Candida from oral lesions of these patients.
Methods and Materials: This descriptive study was performed on 128 patients undergoing chemotherapy in teaching hospitals of Yazd, which was three weeks pass receiving their first medicine. Oral samples were prepared from swabs and then cultured in Sabouraud dextrose agar culture media for evaluation of yeast growth, colonization, and identification of species. Samples were examined under the microscope and recorded. Finally, the data were analyzed by SPSS17 software, Chi-square, and Man-Whitney tests.
Results:128 patients participated in this study, which included 45 males (35.15%) and 83 females (64.85%) with an average age of 40.16 ± 19.95 years. 84 patients (62.65%) had candida in their oral cavity, of which 79 were candida albicans and 5 were Non-albicans Candida. No significant correlation was found between the type of candidates, type of cancer and the frequency of Candida albicans with the age and sex of the patients (P-value <0.05).
Conclusion: Based on the results of this study, the prevalence of Candida albicans in patients undergoing chemotherapy is higher than Non-albicans Candida. Patients with leukemia are more susceptible to Candida infections.

Abstract

Background and objectives: Modeling cancer in vivo is a very important tool to investigate cancer pathogenesis and molecular mechanisms involved in cancer progression. Laboratory mice are the most common animal used for rebuilding human cancer in vivo. Cancer stem cells (CSCs) are the main reason of failure in cancer therapy because of tumor relapse and metastasis. Isolation of cancer stem cells helps us to study their function and behavior. In the current study we separate cancer stem-like cells using sphere formation assay then investigate their tumorigenicity in xenograft tumor model.
Methods: YM1 cancer cells were cultured in serum-free media (SFM) in low adherent culture dishes for enrichment of cancer stem cells. The resulting spheres containing cancer stem-like cells were dissociated into single cells and were injected into the dorsal flank of B6 nude mice.
Results: A few days after injection, subcutaneous tumors formed. The growth curves of the resulting tumors were plotted using their weekly recorded lengths. The tumors' volume and weight were measured. The size of resulting tumors was appropriate to the number of cells injected. Pathological analysis confirmed esophageal origin of the resulting tumors.
Conclusion: Using laboratory mice models is a practical modeling system that provides us investigation of human tumors pathogenesis in vivo.

Abstract

Background and objective: Psychological status is one of the quality of life (QOL) domains which can be affected by hyposalivation caused by head and neck radiotherapy. This study aimed to evaluate the psychological status of head and neck cancer (HNC) patients who were suffering from hyposalivation after at least one week of receiving radiotherapy.
Material And Methods: This descriptive-analytic study was performed on 44 HNC patients with history of hyposalivation caused by radiotherapy, in 5 Azar hospital from 1397-1398. General Health Questionnaire (GHQ-28) was used for data collection. Shapiro-Wilk test, parametric and nonparametric tests and SPSS 18 software were used for statistical analysis.
Results: The average psychological status of patients was 27.50 out of 84 (lower score indicated better psychological status). The mean score of physical health was 7.39, social functioning was 6.16, anxiety/insomnia were 12.64 and depression was 1.33 with the maximum being 21. Psychological status was not significantly different between genders. Ageing caused all subscales to increase except depression. A direct relationship between social functionality and psychological status was reported.
Conclusion: Hyposalivation due to radiotherapy affects psychological status and its components in patients. Also, various factors such as ageing and lower education level can be effective in reducing psychological status in the patients with head and neck cancer who were suffering from hyposalivation due to receiving radiotherapy.

Abstract

Alternative splicing can alter genome sequence and as a consequence, many genes change to oncogenes. This event can also affect protein function and diversity. The growing number of study elucidate the pathological influence of impaired alternative splicing events on numerous disease including cancer. Here, we would like to highlight the significant role of alternative splicing in cancer biology and emphasize the necessity for conducting more research into target alternative splicing as a treatment for cancer.

Abstract

Background and Objective: Cancer is one of the most serious diseases. Doxorubicin is a type of chemotherapy drug used to treat a variety of cancers. Doxorubicin is a type of chemotherapy drug used to treat a variety of cancers. However, its side effects have limited its use. The aim of this study was to synthesize and evaluate polymer micelles containing doxorubicin and evaluate its toxicity on MCF7 breast cancer cells and HepG2 liver cancer cells.
Material and Methods: For this purpose, PBMA-b-POEGMA diblock copolymer was first synthesized using the RAFT method and confirmed by GPC. Dynamic light scattering (DLS) and Transmission electron microscope (TEM) were used to observe the morphology, size, and polydispersity of the micelles. In addition, in vitro cytotoxicity of DOX-loaded polymeric micelles against MCF7 cells and HepG2 cells were assessed. Furthermore, cell uptake and apoptosis assay of DOX-loaded polymeric micelles against MCF7 cells were evaluated.
Results: The TEM image revealed that the nanoparticles were spherical and uniform. The particle size and polydispersity measured by DLS were 35 nm and 0.13, respectively. The drug encapsulation efficiency and drug loading contents were 50±3.46 % and 4.53±0.29 %, respectively. The drug release rate was reported 69% in saline phosphate buffer (pH 7.4) within 24 hours. The results showed that micelles containing doxorubicin had a greater effect on MCF7 cell viability than the free drug. The MTT assay demonstrated that micelles were biocompatible to HepG2 cells while DOX-loaded micelles showed significant cytotoxicity. The IC50 of doxorubicin-loaded micelles against MCF7 cells were obtained to be 0.5 μg/ml. It was further shown that micelles containing doxorubicin had higher cell uptake and apoptosis than free drugs on MCF7 cells.
Conclusion: These polymeric micelles are an ideal candidate to deliver anticancer agents into breast cancer cells.
